All three arachidonate metabolites were generated in small amounts in the lungs as early as 13-15 weeks' gestation. Production of eicosanoids increased during development and peaked in the preterm stage of 22-24 weeks, while the production of PGE2 decreased. The amount of leukotriene generation increased during development. Since eicosanoid generation has been associated with mucus production in epithelia other than the lung, we examined the effects of inhibitors of arachidonate lipoxygenase (phenidone) and 5-lipoxygenase (esculetin) on mucin production in the fetal lung. These two inhibitors reduced mucin production and the overall abundance of mucus-producing cells in the lungs by 43-50%.
